Recursion Announces Phase 2 Data of REC-994, a First-in-Disease Investigational Treatment for Symptomatic Cerebral Cavernous Malformation (CCM), has Met its Primary Endpoint of Safety and Tolerability

REC-994 also demonstrates encouraging trends in objective MRI-based exploratory efficacy measures at the highest dose and the company plans to advance development of REC-994 for the potential treatment of symptomatic CCM in subsequent studies.

SALT LAKE CITY, Sept. 03, 2024 (GLOBE NEWSWIRE) — Recursion (NASDAQ: RXRX), a leading clinical stage TechBio company decoding biology to radically improve lives, today announced top-line results of the SYCAMORE trial, a 12-month Phase 2 randomized double-blind, placebo-controlled, safety, tolerability and exploratory efficacy study for REC-994 in symptomatic CCM patients.

REC-994 met its primary endpoint of safety and tolerability, demonstrating a similar profile across placebo and both 200mg and 400mg dosage-arms with regard to the frequency and severity of adverse events after 12 months of treatment. Magnetic resonance imaging-based secondary efficacy endpoints showed a trend towards reduced lesion volume and hemosiderin ring size in patients at the highest dose (400mg) as compared to placebo. Time-dependent improvement in these trends at the 400mg dose was also observed in this signal-finding study. Improvements in either patient or physician-reported outcomes were not yet seen at the 12 month time point. A meeting with the FDA is anticipated as soon as practical to discuss plans for an additional clinical study. Recursion plans to present data from this trial at a forthcoming medical conference and intends to submit these data for publication in a peer reviewed scientific journal.

“These studies are making significant strides in the development of therapeutics for CCM. The data from this readout is an impressive start and will provide a valuable contribution to the existing CCM literature and strongly supports the need for a future study, with a longer duration and a larger patient cohort,” said Dr. Jan-Karl Burkhardt, MD, Division Head, Cerebrovascular Surgery, University of Pennsylvania and Principal Investigator of the study. Connie Lee, Psy.D., founder and CEO of the Alliance to Cure Cavernous Malformation added: “I speak for the patients who have participated in the trial and those who have been cheering from the sidelines while waiting for news. This promising start is a critical step forward and will bring hope to thousands of families who currently have no options but brain or spinal cord surgery. The Alliance to Cure Cavernous Malformation looks forward to partnering with Recursion as they move to the next stage of the REC-994 program.”

“We are encouraged by the recent data from our signal-finding Phase 2 study in CCM, where the trial successfully met its primary safety endpoint and became the first investigational therapy to demonstrate safety alongside some promising trends in exploratory efficacy endpoints. These results provide critical insights that will inform our next study design, including exploring study duration, higher doses, and a larger cohort of patients,” said Najat Khan, Ph.D., Chief R&D Officer and Chief Commercial Officer of Recursion. “This is the first of several key clinical readouts for the company and represents an early proof-of-platform milestone for our constantly evolving Recursion OS, as we build upon our success in drug discovery with expertise and execution in mid-phase development. We are deeply grateful to the patients and investigators, and we are committed to advancing potential transformational therapies for CCM and beyond.”

Background on Cerebral Cavernous Malformation (CCM)

CCM is a neurovascular condition that impacts approximately 360,000 symptomatic individuals in the US and EU5. The disease is often underdiagnosed and potentially affects over 1 million patients worldwide. CCM manifests as vascular malformations of the spinal cord and brain characterized by abnormally enlarged capillary cavities without intervening brain parenchyma. Patients with CCM lesions are at substantial risk for seizures, headaches, progressive neurological deficits, and potentially fatal hemorrhagic stroke. Currently, only non-pharmacologic treatments including microsurgical resection and stereotactic radiosurgery are available options for this high unmet need patient population. However, surgical resection or stereotactic radiosurgery is not always feasible because of location and may not be curative.

About REC-994

REC-994 is an orally bioavailable, superoxide scavenger small molecule under development for the treatment of symptomatic CCM. The potential of REC-994 in CCM was demonstrated using the earliest version of what would become the foundational technology underlying the Recursion OS. Subsequently, REC-994 demonstrated preclinical activity in models for CCM and tolerability and suitability for chronic dosing in Phase 1 single ascending dose escalation (SAD) and multiple ascending dose escalation (MAD) trials in healthy volunteers directed and executed by Recursion. Recursion has sought and received Orphan Drug Designation for REC-994 in symptomatic CCM in the US and Europe.

About the Trial

Our Phase 2 SYCAMORE clinical trial is a randomized, double-blind, placebo-controlled study of two doses of REC-994 in participants with CCM. The primary endpoint of the study is safety and tolerability. Secondary efficacy endpoints include MRI-based endpoints, clinician and patient reported outcomes, as well as selected biomarkers. This trial was fully enrolled in June 2023 with 62 participants, and 80% of participants who completed 12 months of treatment have entered the long-term extension study. This signal-finding study was not powered to demonstrate statistical significance.

Saudi Red Sea Authority Launches More Than a Sea Campaign

The Saudi Red Sea Authority (SRSA) has launched a campaign titled "More Than a Sea," which revolves around four main pillars of SRSA's roles: regulations, sustainability, attracting investments, and promoting tourism experiences in the Red Sea, to highlight its contributions toward building a thriving coastal tourism sector. The campaign, rooted in SRSA's duties, focuses on its regulatory role in setting policies, strategies, plans, programs, and initiatives necessary for regulating navigational and marine tourism activities, issuing licenses and permits, and preparing the infrastructure for these activities. Additionally, SRSA has issued seven regulations in collaboration with the relevant entities to regulate coastal tourism activities in Saudi Arabia. More Than a Sea also addresses one of SRSA's roles of attracting investment in the Red Sea, which offers a promising investment environment in the region with its unique features, such as a population of around 7 million people supporting coastal tourism along an extensive 1,800 km coastline, diverse landscapes, climates, cultures, heritage, valuable marine treasures, charming nature, creating an astonishing environment for enthusiasts and practitioners. The campaign also highlights SRSA's efforts to promote various coastal tourism activities, such as cruise ships, yachts, diving, snorkelling, recreational fishing, recreational boats, and beach activities. It encourages tourists and visitors to explore and live a unique coastal experience in the Red Sea. When it comes to sustainability, More Than a Sea emphasizes SRSA's objectives toward environmental protection by leading the efforts with relevant entities from the public and private sectors to develop a mechanism to ensure protection of marine environment, fostering the growth of the blue economy, preserving natural assets and pristine resources in the Red Sea, as well as contributing to the production of nautical maps that define safe routes and protect coral reefs, along with managing marine waste, in stalling mooring buoys, and establishing weather monitoring stations. The Red Sea coast features over 150 beaches, over 1,000 islands, and 130 cultural, historical, and biological assets. It is rich in natural beauty, enchanting treasures and wonders, over 20 blue holes, over 500 diving sites, and diverse culture and heritage, including traditions, customs, architecture, clothing, and over 50 traditional dishes. In line with Saudi Vision 2030's objectives to diversify income sources, SRSA's goals for the coastal tourism sector are to contribute SAR85 billion to the GDP by 2030, increase spending to reach SAR123 billion and create 210,000 jobs during the same period, thereby diversifying Saudi Arabia's non-oil income sources. LMRA implements new reconciliation procedures for work permit violations

Manama, The implementation of Edict (12) of 2024, which amended Article (40) of Law (19) of 2006 regarding the regulation of the labour market, Nibras Mohammed Talib, CEO of the Labour Market Regulatory Authority (LMRA), announced. This amendment focuses on modifying the procedures and criteria for reconciliation in offences related to work permits. The LMRA remains committed to continuously reviewing and improving the mechanisms and procedures governing the labour market, Talib said. He added that these amendments align with the National Labour Market Plan (2023-2026) and aim to enhance compliance with laws and regulations. The amendments resulted from continuous consultations with stakeholders, which would provide employers with more flexibility in resolving violations through reconciliation, particularly in cases where an expired work permit is detected for a foreign worker employed by the same employer. Furthermore, the LMRA clarified that it consistently reviews and updates its procedures and regula tions to ensure alignment with labour market developments, thereby enhancing Bahrain's competitiveness and supporting the sustainability of the commerce sector, which contributes to the Kingdom's progress regionally and internationally in this sector. According to the Edict, reconciliation is permitted for the offence stated in paragraph (b) of Article (23) of the Labour Market Regulation Law, which prohibits employers from employing a foreign worker without a valid work permit, by paying a reconciliation fee of BHD 500. In cases of repeated offences, the reconciliation fee increases to BHD 1,000. The LMRA further explained that if an employer is found employing a foreign worker whose work permit has expired, the reconciliation fee will be staggered: BHD 100 if the violation is detected within the first ten days after the permit's expiration, BHD 200 if detected after ten days but before twenty days, and BHD 300 if detected after twenty days but before thirty days. After thirty days, the minimum statutory f ine for the offence applies. Additionally, the amendments to the LMRA Law allow reconciliation for the offence stated in paragraph (a) of Article (23), which prohibits foreign workers from working in the Kingdom without a valid work permit, with a reconciliation fee of BHD 500 for first-time violations. These amendments aim to provide employers with flexibility in rectifying their situations in cases of non-compliance with work permit procedures or renewals, encouraging adherence to laws and regulations, and reducing the incidence of illegal employment. Employers are also given the opportunity to rectify the status of their workers by obtaining the necessary permits, contributing to the sustainability of business operations. The amendment also extends the reconciliation period from seven to 14 working days to facilitate the rectification process. The LMRA highlighted that employers have the option to renew work permits before they expire, which helps avoid violations outlined in the law. The authority has implemented mechanisms to remind employers six months before the permit's expiration, with additional reminders three months and one month prior, as well as on the expiration date itself. The LMRA also contacts employers directly before the permit expires and encourages them to update their contact information to receive all notifications and updates through the Expatriate Management System (EMS).
